SleepDelta Sleep-Inducing Peptide
Overview
DSIP is a naturally occurring nonapeptide first isolated from rabbit cerebral venous blood in 1977 at the University of Basel, named for its ability to increase slow-wave (delta) EEG activity — the deepest, most restorative sleep stage. Subsequent research revealed a far broader profile spanning stress, pain, hormone regulation, and antioxidant activity. Unusually, its gene has never been identified. It crosses the blood-brain barrier, which distinguishes it from most peptides and underpins its central effects.
Mechanism of Action
DSIP acts as a neuromodulator rather than a sedative, normalizing dysregulated sleep architecture and increasing delta-wave activity. It reduces basal ACTH and blunts stress-induced cortisol elevation (HPA axis modulation), shares partial structural similarity with dermorphin and is hypothesized to have partial opioid receptor agonism (mild analgesia, attenuation of alcohol and opiate withdrawal), scavenges reactive oxygen species and protects mitochondrial function under hypoxia, and stimulates GHRH and GH release while inhibiting somatostatin — producing a net pro-GH environment. Anticonvulsant activity has also been documented in animal models.

Daily SC 30–60 minutes before bed: 300 mcg (wk 1–2) → 500 mcg (wk 2–4) → 1,000 mcg (wk 4+ maintenance). Cycle 8–10 weeks on / 4–6 weeks off, or a 5-days-on / 2-days-off weekly structure. See the full DSIP protocol at /protocol/dsip.
Reconstitution Guide
Vial size: 10 mg
Water volume: 1 mL bacteriostatic water
Concentration: 10 mg/mL (10,000 mcg/mL) — 100 mcg per unit
Add 1 mL of bacteriostatic water slowly along the vial wall — do not shake. 300 mcg = 3 units, 500 mcg = 5 units, 1,000 mcg = 10 units. Use a 29–31 gauge insulin syringe for precision at these low volumes.
Storage Guidelines
Before reconstitution: Store lyophilized powder frozen or refrigerated, protected from light.
After reconstitution: Refrigerate at 2–8°C. Stable up to 28 days with bacteriostatic water. Do not freeze.
Half-Life & Stability
Half-life: ~30 minutes in plasma — short, which is why dosing is daily and timed 30–60 minutes before bed
Stability: Low molecular stability compared with most peptides — source quality and cold-chain handling matter significantly. Inspect solution before each use. Effects can attenuate with continuous daily use; cycle 8–10 weeks on / 4–6 weeks off to preserve responsiveness.
